A fascinating study from @MizLabUT using CRISPR screen revealed that loss of purine and pyrimidine enzymes activates autophagy. Interestingly, purine starvation induces autophagy through mTORC1 while pyrimidine depletion does not. @Gerta_Hoxhaj @Manning_BD

In addition to SREBP, MYC, and HIF1 now ATF4 can be considered as a canonical target of mTORC1 signaling downstream of growth signals. The icing on the cake, the mTORC1-ATF4 axis stimulates cystine uptake and glutathione synthesis. Congrats @Manning_BD and @meghan_torrence .

A very elegant piece of mechanistic research demonstrating that proliferating cells carry out aerobic glycolysis when the NAD+ demand for pyruvate oxidation reactions surpasses the demand for ATP. Consequently, the ETC cannot regenerate NAD+ leading to Warburgian metabolism.
