Big news: My Starting Grant was FUNDED, so my group is moving to ETH Zürich @ETH_en in 2023 where we will continue to study the molecular biology of cool new phages. We will be based at D-HEST together with our friend @MartinLoessner and his group. Great times ahead 🍾💰🧫! 1/2

Achievement unlocked - first phage isolated that has no relevant similarity to anything previously known (and the genome is big, >>100kb!). Best whole-genome BLAST hit has 0% coverage / 70% identity due to a short 100bp stretch of homology in a tail gene. Exciting! 😁🧬🔥

Bacterial immunity is so satisfying to watch 🦠🛡️! The plates below show dilutions of twelve phages on a reference host (left) vs. some exciting anti-phage defense (right). No shaky ambiguous phenotypes like in some other areas of microbiology 😅...

Totally in love with these phages (new and undescribed group infecting E.coli) that form a "crown" of clear lysis around their plaques. Exciting! 🤩🧫 The weird thing is that these "crowns" are discontinuous with the plaques. Anybody ever saw anything like that or has ideas?

These phages here confirmed once more that "trying out various things which vaguely make sense" is the superior problem-solving strategy in molecular biology... too often there are so many unknown parameters that one can simply not build a rational road to the goal. 🛠️🧫😅

Last week I visited a Pestalozzi school camp and discussed with the kids about phages in environment and clinics. Given the camp's aim of linking science + art, we also colorized some TEM pictures. I was stunned by the kids' individualism and creativity! Example before / after 👇🏼

With our high schoolers today: plasmids and phages. The F-plasmid defends E. coli against phage T7 (see F+ vs. F-) but it sensitizes cells to phages M13 and MS2 that bind its sex pilus ⚔️🧫. T4 doesn't care 😁. Great discussions how good and bad are often context-dependent!

We finally completed our arrival in the new institute building because the students now customized all benches with black cover. I don't know why white benches are standard, but everything - colonies, plaques, even microscopy slides - are much better visible on a dark background.

Fun observation of the day: The virulent phage in the second lane clears the turbid plaques of the phages in the adjacent lanes. I suspect very potent secreted lysins 🤔🧫. (these are a couple of new / unpublished phages on E. coli K-12)

Today was day 1 in the last year of my expiring junior PI position at @biozentrum - lots of exciting science with my student dream team and great colleagues, but all good things eventually end. Nobody knows what the future holds though I will for sure miss the view. 🧳🌆🌄

Temperate phages (forming prophages) often have light turbid plaques like these since lysogens are self-immune. See the one clear plaque of an obligately lytic mutant - it's the reason lambda's iconic lysogeny genes are cI, cII, etc. - c for clear plaque. Exciting in real life 👍🏼

I think we finally got blue / white selection with lytic phages to work also in our hands (lacZ alpha in phage genome, lacZ omega in host, X-gal in top agar)... 🔵🔵🔵🤩 This was clearly commonly used in the old days but somehow it took us a while to get the conditions right. 🤔

Well, that was easy - at least if the bacterial pathogen is not an obscure organism, we usually find lots of phages (here: uropathogenic E.coli). The more tricky next steps would be 1) to choose the right ones for therapy and 2) to effectively apply them to the patient... 🧬💉💊

Today I learned: The flagellar serotype "H" of enterobacteria stands for German "Hauch" ("breath") because swarming colonies on agar look like when you breathe at a glass slide... and the "O-antigen" was first seen with non-motile strains ("ohne Hauch"). I am stunned... 🤔🌬️🧫

Deeply touched by a surprise dinner yesterday with all lab members and alumni to celebrate the (re)birth of our group. They composed a wonderful book about our history with touching entries from many friends and colleagues... I am speechless 🙏🏼❤️.

#PhageOfTheWeek is back with phage P2! This virus does a rather bad job as a predator (those plaques! 😁), but that’s okay – as a temperate phage it doesn’t need active infections very often. Instead, it elegantly boosts lysogen fitness through bacterial immunity 🦠🛡️. A 🧵 1/15

#OnThisDay three years ago during a May Hike I found a huge pile of rotting plants behind Hörnli cemetery in Basel. Some smelly juice ended up in the lab where Enea @phagemuffin eventually isolated phage Paride from it - now a major focus in the group! 😁♻️🧪

Phishing new phages never gets old - first time in the new lab. 😁🧫🎣 (direct isolation from river water on a usually difficult E. coli host - I guess abundance and diversity might be favored by all the materials carried in by current high water)

Today just out of a mood I asked our son if he is German, Swiss, or (like his beloved grandma) Greek. He evidently thought that's a stupid question and answered "Papa, I am Johann!" His mother and I count this as a parenting win 🌍☃️👍🏼.

#PhageOfTheWeek is back with a little monster that started as a lab pet – temperate phage phi80, a cousin of phage lambda. We crossed paths when I was a PostDoc and then things escalated quickly, causing great relief for some people and significant grief for others. A 1/12
