Went with my partner upstate and the AirBnB host's cat took us for a guided hike along the Appalachian trail. Apparently this is what she does with every guest. She would complain when we took a wrong turn off the trail and knew the way back. Amazing cat. Would apocalypse with.

I love my lil hand made agarose gel transilluminator. Really proud that I circumvented a really shitty patent essentially owning the idea of "perpendicular illumination". Well, it's all freehand and no PCBs or glass lenses so take a hike. Will make for anyone. $64 + shipping!

Still very much in disbelief that two degreeless heathens in a home lab with no formal training managed to produce a reference quality bacterial genome of a really hard-to-crack microbe and that our assembly is now THE representative genome of that organism. Amateur biology FTW!

Mmmm, future tech. I got these cool sodium ion batteries in 18650 size for about $1.50/piece. Rated at 1300mAh and 3.90Wh. Nominal voltage of 3.0v. Extremely cold resistant. No thermal runaway. Far more sustainable. Excited to design using salt-tek, lol.

Since my big autoclave died, I've been using this InstantPot pressure cooker exclusively for all my sterile work. Has not given me contam once and I often run various antibiotic-less media. I set it to max temp n pressure. Set to Pressure Cook mode 30mins.

Trying a quick gel purification trick I saw in a paper recently (reference needed). Poke 0.5mL tube with 22 guage needle. Stuff with ~50uL of cotton. Tamp it down. Wash with 100uL dh2o. Keep collection tubes for reuse. Autoclave washed tubes. Load gel slab. Spin 14k 10mins. Done.

Nine PCR optimization runs later and I got my bands. DMSO + very specific annealing temp + Taq. Q5 and PrimeStar failed miserably. Some slight secondary amps, but whatever. Biology is hard.

I love these autoclavable "petri dishes". They are 50mm bead boxes used for storing fly fishing tackle, but are made of polypropylene and have snap fit lids. Been using these for 3 years now and never turning back. Yay, minimizing disposable plastic waste!

Finally. Toy-grade PCR machines that actually work. So tired of the pretentiousness surrounding thermocyclers. It's a hot-cold-hot machine. Trivial with modern materials. No reason it should ever be so expensive. Ever. Thus one retails for $245.

I love this stuff! Becomes like putty in boiled water and then stiffens rock hard as it cools. Excellent manual sculpting material and infinitely reusable. Good machinability too. Scraps can be melted together. Takes dyes well too. Cheap.

#BLUESOUP LETS GOOOOOOOOO Plated on King's Agar A and B (pyocyanin and fluorescein ind respectively) and will incubate at 30°C for 24 to 48hrs. Resuspended in LB and then serially diluted in 10x steps to 10,000x. Plated 10uL per dish. Rest stored in back of fridge at 4°C. 1/2

Alright, we got another pilot #BlueSoup experiment underway to test if the glowy bugs make blue on soft cheese! A few articles suggest that Pseudomonas fluorescens can make cheese blue so I put some mozz in 12 well plates + 1mL of mozz broth + 30uL of BS1-A cells. 🧵1/4

Finally, I finished drafting my take on a digital firefly. About the size of a large adult, this small circuit is solar powered (charges a tiny supercapacitor) and flashes at dusk for a minute or two...forever. Tricky to route but programmable via ATTiny10 and no lithium batts!

@timrpeterson Biology lags behind software because biology is not dead silicon that remains unchanged, where the parts are discrete and fully defined, where whole systems are simulatable and the iteration cycle is seconds not years. Software ≠ hardware ≠ wetware. Myopic is an understatement.

One thing I ask if you use my chromoprotein plasmids in educational settings is to remind your students that these genes came from our oceans, gifts from the great reefs, and it's up to us to protect these envrionments. Keeping our oceans healthy is of the utmost importance. 🌊🙏
