Overall we establish NAD+ depletion as a shared mechanism by SIR2 defense systems to protect bacteria against phage! NAD+ seems to be a central molecule in the phage-bacteria interface. Excited to see what comes next :)

Finally, we show other systems where SIR2 domains deplete NAD+ and contribute to an abortive infection phenotype, and one system where NAD+ depletion and phage protection does not involve culture collapse.🧐

How did we find these proteins? We used a “phage mating” technique promoting genetic exchange between phages that are sensitive to DSR2 and phages that are not. Genome analyses of hybrid phages enabled us to pinpoint the exact phage proteins that activate or repress DSR2!😎

We show that DSR2 recognizes newly translated phage tail tube proteins to become an active NADase. In addition, we found that some phages in this family carry a small protein, DSAD1, which binds and inactivates DSR2.

Recently, many anti-phage defense systems were shown to contain SIR2 domains. One of them, described by Gao et al, is DSR2, a single-gene defense system with a SIR2 domain at the N terminus. But how do DSR2 and other SIR2 systems defend against phages? #WhatIsTheMechanism ?
